Breast Cancer

T-DM1 Found to Prolong Progression-Free Survival in Patients with HER2+ Metastatic Breast Cancer

The experimental antibody-drug conjugate trastuzumab emtansine (T-DM1) has shown significant improvement in progression-free survival (PFS) compared with capecitabine plus lapatinib among patients with HER2+ locally advanced or metastatic breast cancer (MBC).

Weekly Paclitaxel After Adjuvant FAC Offers Modest, but Clinically Relevant Improvement in Disease-Free Survival in Patients with High-Risk, Node-Negative Breast Cancer

Adjuvant therapy including 5-fluorouracil, doxorubicin, and cyclophosphamide (FAC) followed by weekly paclitaxel was associated with a slight but significant improvement in disease-free survival.

Updated, Subgroup Analyses of BOLERO-2 Confirm Efficacy of Everolimus plus Exemestane in Postmenopausal Women with Advanced Breast Cancer

Several analyses of the international randomized phase 3 Breast Cancer Trials of Oral Everolimus-2 (BOLERO-2) plus exemestane versus exemestane alone for the treatment of postmenopausal women.

First-line Lapatinib/Taxane Found to be Inferior to Trastuzumab/Taxane in Patients with HER2+ Metastatic Breast Cancer

Taxane-based chemotherapy combined with lapatinib results in shorter progression-free survival (PFS) than with trastuzumab as first-line treatment for patients with HER2+ metastatic breast cancer.

Leukemia

Ponatinib Active in Patients with CML and Ph+ ALL Resistant/Intolerant to Dasatinib or Nilotinib or Who Have the T315I Mutation

The potent, oral, pan-BCR-ABL inhibitor ponatinib has demonstrated robust antileukemic activity in a heavily pretreated population of patients with chronic myelogenous leukemia or Ph positive (Ph+) acute lymphoblastic leukemia who are resistant or intolerant to dasatinib or nilotinib or have the T315I mutation.

Lung Cancer

Crizotinib Provides Clinically Meaningful Antitumor Activity in Patients with Previously Treated ALK-Positive Advanced Non–Small-Cell Lung Cancer

The first-in-class, oral, potent, and selective small-molecule competitive inhibitor of anaplastic lymphoma kinase (ALK), crizotinib, was found to have a durable overall response rate and a median progression-free survival of 8.1 months in patients with ALK-positive non–small-cell lung cancer (NSCLC).

Multiple Myeloma

Pomalidomide Active in Patients with Multiple Myeloma Refractory to Lenalidomide, Bortezomib—or Both—and in Transplant Failures

Pomalidomide in combination with low-dose dexamethasone has shown a 30% overall response rate in patients with multiple myeloma refractory to lenalidomide, bortezomib, or both, including those with prior transplant.

Carfilzomib, Lenalidomide, Low-Dose Dexamethasone Provides Rapid, Deep Responses in Patients with Newly Diagnosed Multiple Myeloma

The combination of carfilzomib, lenalidomide, and low-dose dexamethasone has been shown to be highly active in patients with newly diagnosed multiple myeloma and provides rapid and deep responses.

Palliative Care

Early Palliative Care Intervention Improves Satisfaction with Care, Prevents Decline in QOL in Patients with Metastatic Disease

Patients with metastatic cancer who received early palliative care intervention had immediate improvement in satisfaction with care and later improvement in quality of life (QOL) and symptom control compared with standard care.

Prostate Cancer

Continuous—vs Intermittent—Androgen Deprivation Extends Overall Survival in Men with Minimal Disease Hormone-Sensitive Metastatic Prostate Cancer

Continuous androgen deprivation may be a preferred treatment option for hormone-sensitive metastatic prostate cancer in men with minimal disease spread, according to results of a long-term international phase 3 trial reported at the 2012 Annual Meeting of the American Society of Clinical Oncology.

Interim Analysis Shows Abiraterone Acetate Significantly Improves Radiographic Progression-Free Survival in Chemo-Naïve Metastatic Castration-Resistant Prostate Cancer

Patients with asymptomatic and mildly symptomatic chemotherapy-naïve metastatic castration-resistant prostate cancer had significantly improved radiographic progression-free survival (rPFS) and showed a strong trend for increased overall survival (OS) following treatment with abiraterone acetate plus prednisone compared with prednisone alone.
